I'd give Jamal Murray the edge defensively. His superior size (well, bulk) allows him to switch onto bigger players better than Herro. Able to physically fight on opposition drives. Able to play long minutes at SG alongside another small guard. More versatile defender.
The main issue is Jamal Murray is a proven playoff performer while Herro has yet to have a strong playoff showing. He has struggled in the past in the playoffs but this is breakout year. It looks like Herro has turned that corner and become a legit star in his own right.
