I'm skeptical it wasn't just a missed assignment rather than actually misreading which way they were going...

I understand what you're thinking here, and I wondered the same, but on the replay you can see that just about every Hornets player was standing on the right-hand side of the Lakers players, which you would only do if you were defending the basket at that end of the court. All Kobe had to do was take off sprinting (he was standing on the left-hand side of the group already, with no one to his left) and it was an easy basket.
